The game takes place before the events of the first Serious Sam game. Crotein, the leader of the Jerusalem Brotherhood, has stolen an ancient artifact known as the "Tear of Isis". With this powerful relic, Crotein plans to conquer the world and destroy all non-believers. Serious Sam, the galaxy's most renowned bounty hunter, is tasked with stopping Crotein and his minions.
